| FoldHelper™-905c is a sextuple recombinant baculovirus, which provides for the expression of human molecular chaperones Hsp90 (Acc# AAH12807), Hsp40 (Acc# BAA02656), Hsc70 (Acc# AAK17898), Hop (Acc# NP_003923), p23 (Acc# AAA18537) and a GFP reporter (Acc# AAA58246). All proteins are found in soluble protein fraction and could be detected as distinct bands after separation in SDS-PAGE and staining with Coomassie blue (Fig. 1 in Multi-chaperone systems). Molecular chaperones expressed by FoldHelper™-905c can dramatically improve yield of Hsp90-client proteins, such as steroid receptors (Fig. 2 in Multi-chaperone systems). | ||
| Hsp90 is a molecular chaperone which facilitates folding of a large number of therapeutically relevant proteins, such as steroid/nuclear receptors, Hsp90 client protein kinases, and viral polymerases. A comprehensive list of Hsp90 client proteins could be found on Prof. Picard's lab web site http://www.picard.ch/downloads/Hsp90interactors.pdf. Hsp90 works in ensemble with several other proteins. Hsp40, Hsp70/Hsc70, Hop, and p23 are required in addition to Hsp90 for activation of steroid receptors (Pratt W.B. and Toft D.O, Exp Biol Med, 228:111-33, 2003; Pratt W.B. et al., Handb Exp Pharmacol., 172:111-38, 2006). Another molecular chaperone, Cdc37 could be required in addition to Hsp90, Hsp40, Hsp70/Hsc70, and p23 for activation of Hsp90-dependent protein kinases (MacLean and Picard, Cell Stress Chaperones, 8:114-9, 2003). Cdc37 can be provided using a separate recombinant baculovirus FoldHelper™-37, which enables its expression to a high level. | ||
