As documented in more than 100 studies, N- or C- terminal fusions of target proteins with a maltose binding protein (MBP) tag can increase yield and solubility of the protein product. Furthermore, the MBP tag allows for affinity purification of the fusion product on the amylose resin. The MBP tag is sometimes combined with a 6xHis tag as the latter is more widely used for affinity purification. We provide vectors for the expression of target proteins with either an N-terminal MBP tag (pAB-MBP™) or an N-terminal 6xHis-MBP tag (pAB-6xHis-MBP™). In either case, the N-terminal tag can be cleaved from the fusion protein with thrombin. A versatile pAB-6xHis-MBP™ vector could be also used to provide the MBP tag on the C-terminus of the target protein.
MBP tag vectors can be combined with any of the listed below linearized baculovirus DNA vectors, i.e. ProEasy™, ProGreen™, ProFold™-ER1, ProFold™-C2, ProFold™-PDI, and ProFold™-0.
