  Most insect media can be used to overlay monolayers during transfection as long as it does not contain serum or antibiotics, both of which reduce transfection efficiency. Synthetic serum-free media also contain relatively high concentrations of lipids which may interfere with transfections. Transfection conditions should therefore be optimized if you decide on a medium other than that recommended in this tutorial. We routinely use TC-100 medium (Sigma, Cat# T3160) not supplemented with serum or antibiotics because of its lowest lipid content. The transfection protocol described in this tutorial was optimized with this medium. Synthetic Insect-xpress (Lonza Cat# BW12-730F) medium could be used with the same protocol, and some customers used SF900 media from Invitrogen. A side by side analysis for transfection efficiency has not been done.
