A need to express several proteins in the same cells exists if the proteins have to interact to form a functional multi-protein complex or are part of an enzymatic cascade.
pAcAB3 plasmid transfer vector can provide a high level of expression for up to 3 target proteins using the same recombinant baculovirus. Three genes encoding proteins of interest are sequentially inserted into unique sites of the vector to be expressed under the control of strong, very late baculovirus promoters. This takes more time than simultaneous construction of 3 single recombinant plasmid constructs. However, it pays off as performance of the triple recombinant baculovirus far exceeds performance of 3 single recombinant baculoviruses co-infecting host cells. Click on TECHNOLOGY for details.
Insertion of 3 genes into the pAcAB3 vector requires only basic E.coli cloning skills. The next step, co-transfection of the resulting triple recombinant plasmid construct with linearized baculovirus DNA is also routine and the same as with other plasmid transfer vectors.
pAcAB3 plasmid transfer vector can be combined with any of the listed below linearized baculovirus DNA vectors, i.e. ProEasy™, ProGreen™, ProFold™-ER1, ProFold™-C2, ProFold™-PDI, and ProFold™-0.
